by: Dr. Stephanie Rudolph Bio: Alexander Jackson, PhD is an Associate Professor in the Department of Physiology and Neurobiology at the University of Connecticut (UConn) in Storrs, CT. Following a BSc in Physiology (McGill), he pursued a PhD in Neuroscience (Harvard) with Bruce Bean, studying subthreshold ionic currents underlying spontaneous firing in hypothalamic pacemaker neurons. He subsequently pursued a postdoctoral fellowship with Roger Nicoll (UCSF) studying glutamate receptor trafficking and synaptic transmission. With support of a K99 (NIMH), he received further training with Luis de Lecea (Stanford), studying neurotransmission in hypothalamic neural circuits. He performed his PhD work with Wade Regehr at Harvard on synaptic transmission in the cerebellum, and his postdoctoral work with Bernardo Sabatini at Harvard on subcellular Ca2+ signaling in the striatum. He currently studies neurons, synapses and circuits in the prefrontal cortex, thalamus, striatum, and related brain regions. These diverse networks contribute to high-level brain function, including cognitive and motivated behaviors. Dysfunction of these areas is also linked to disorders such as schizophrenia, anxiety, and drug addiction. The Carter lab broadly studies how different populations of neurons communicate in their local and long-range networks. They also examine how specific neurons and synapses are regulated by dopamine and other neuromodulators. Lastly, they determine how these distinct circuits are engaged during rewarding and aversive behaviors. Their experiments use a powerful combination of electrophysiology, two-photon microscopy, and optogenetics, both in vivo and ex vivo. How do experiences modify behaviors? How do similar experiences elicit drastically different behavioral responses in the healthy and disease states? These are some of the questions that drive the research in my lab. We particularly focus on the mechanisms underlying the reconfiguration of neural circuits following sensory and behavioral experiences that leads to behavioral adaptation as well as functional maturation during development. While we know in many cases the brain regions that are involved, it remains challenging to identify the neurons that encode the information and the particular information that is processed. To overcome these challenges my lab has developed a new set of genetic tools that have enabled us to genetically identify neuronal ensembles activated by sensory and behavioral experiences, uncover essential circuit components involved in different aspects of learning and memory, and explore the mechanisms by which learning is specifically coupled to synaptic changes on these ensembles to achieve behavioral adaptation. One aspect of CNS development that is largely unknown is how the dynamics of chromatin modifications change during neuronal differentiation. To analyze dynamic changes in the chromatin landscape that regulate gene expression during neurogenesis, migration and circuit formation, we are studying cerebellar granule neurons. These studies show that H3K4me3 and H3K27me3 histone bivalency exists in individual developing granule cells and regulates the timing of granule cell differentiation and glial-guided migration. One of the key cerebellar genes involved in granule cell development and migration, Astn2, has been implicated in autism spectrum disorders. Using a mouse model for loss of Astn2 function, we find that mutant animals have pronounced ASD-like behaviors, including deficits in ultrasonic vocalization, hyperactivity, anxiety and repetitive behavior phenotypes, but do not have motor impairments. A reduction in Purkinje cell spine density and increase in Bergmann glial volume, as well as electrophysiological changes in the cumulative fractions of spontaneous EPSC interevent intervals, suggest that changes in the cerebellar circuitry underlie these non-motor function deficits. Here I will discuss new and unpublished data from our lab on when, where, and how oxytocin is released from hypothalamic neurons to enable maternal behavior in new mother mice. I will focus on maternal responses to infant distress calls, and how oxytocin enables rapid neurobehavioral changes for dams and alloparents to recognize the meaning of these calls. We have built a new system combining 24/7 continuous video monitoring with neural recordings from the auditory cortex and oxytocin neurons of the hypothalamus in vivo. With this documentary approach, we have identified behaviors of experienced and naïve adults learning to co-parent together which also activate oxytocin neurons. I will discuss circuits routing sensory information to oxytocin neurons leading to oxytocin release in target areas important for maternal motivation. Finally, I will discuss longer-term behavioral monitoring over months, examining how single mothers build nests to help ensure pup survival or how this sometimes goes awry. When a cue later occurs without punishment, this unexpected omission of aversive outcome is encoded as reward via activation of reward-encoding dopaminergic neurons. How such activation occurs remains unknown. Using real-time in vivo functional imaging, optogenetics, behavioral analysis and synaptic reconstruction from electron microscopy data, we identify the neural circuit mechanism through which Drosophila reward-encoding dopaminergic neurons are activated when an olfactory cue is unexpectedly no longer paired with electric shock punishment. However, while the flow of information through a neural circuit is digital (mediated by action potentials), the modulation of how information flows through a neural circuit is not. My laboratory’s research seeks to identify the organization, function, and therapeutic potential for ion channels and their binding partners for regulating synaptic function. Ion channel research was propelled by the invention of glass electrodes to precisely measure electrical signals in cell membranes. However, the average neuron in a human brain sends electrical signals across axons that are only 1/1000th the width of a human hair while also sometimes spanning tens of centimeters in length. This precludes the use of standard electrophysiology. Our group has devised and refining optical techniques to measure ion channel activity using genetically encoded optical indicators to overcome these size barriers in the axon and terminals. In this talk, I will describe two new roles of voltage-gated potassium channels and novel binding partners for regulating synapse function using quantitative optogenetic reporters.
