Overview
The Transgenic Facility provides a wide array of services to the college: pronuclear injection, lentiviral transgenesis, rederivation, sperm/egg/embryo cryopreservation and in-vitro fertilization.
Investigators design and assemble a fusion gene construct, submit it to the facility and we will inject the DNA into zygotes. The background strain typically used is FVB, but investigators wishing to use a different strain must discuss the possibility with Dr. Ken Chen beforehand.
Pups are born 19-20 days post-implantation and are housed in the facility animal room until weening age. Candidate pups are then transferred to the respective investigator's animal room through Institute of Animal Studies.
Scheduling
The facility operates on a first come - first served basis. Typically investigators can expect to have their constructs injected within two weeks upon delivery of their construct. Please contact Ken Chen to discuss your project specifications and arrange scheduling. Project requests should be made via the forms in the iLab Core Management software.
Deliverables
The entire process, beginning with the delivery of the construct, to receiving candidate mice resulting from the injection. takes approximately two months. Facility policy is to provide 3 founders or 60 candidate mice, whichever comes first.
Investigator Responsibilities
Recipient labs are required to do southern blot and provide evidence (or lack thereof) of positive mice within four weeks of receiving candidate mice. Failure to do so will result in full cost of reinjection.
