Professional Interests
The invariant component of the SWI/SNF chromatin remodeling complex INI1/SMARCB1/hSNF5/BAF47 is involved in different cellular functions, like, transcription, cell cycle regulation, development, and tumor suppression. Host INI1 and the viral HIV-1 integrase (IN) are required at different stages of HIV-1 infection. The Rpt1 domain of INI1 binds to the C-terminal domain of HIV-1 IN. In fact, INI1-Rpt1 and HIV-1 TAR RNA compete with each other for HIV-1 IN binding as evident from the common residues involved in both these biomolecular interactions. Interestingly, the three-dimensional structure of INI1-Rpt1 domain, despite being part of a protein, shares a close resemblance with HIV-1 TAR RNA. This structural semblance between a host protein and the viral RNA is rather intriguing and raises several interesting questions regarding the host-pathogen cross talk during HIV-1 infection. We are interested in understanding the rationale behind this evolutionary arrangement for a possible modulation of the interaction between the human host and HIV-1 virus, and in the process, identify potential anti-HIV therapeutic targets.
