Focalización de la transducción de señales en neoplasias hematológicas:
Las citocinas desempeñan papeles importantes en la regulación de la hematopoyesis normal y se requiere un equilibrio entre las acciones de los factores de crecimiento hematopoyéticos y los factores mielosupresores para la producción óptima de diferentes linajes de células hematopoyéticas. Los sÃndromes mielodisplásicos (SMD) son conjuntos de enfermedades hematológicas heterogéneas caracterizadas por citopenias refractarias debido a una hematopoyesis ineficaz. Estos trastornos preleucémicos son causas comunes de anemia en los ancianos y su incidencia está aumentando rápidamente. También hemos demostrado que las vÃas de la superfamilia TGF-beta/smad, IRAK y MAP quinasa están sobreactivadas en los SMD. Nuestro trabajo ha dado lugar a ensayos clÃnicos con Luspatercept, Galunisertib, CA4948, ARRY-614 y varios otros agentes prometedores en los SMD. Estamos tratando de estudiar los mecanismos moleculares que conducen a la activación de estas vÃas en los SMD y estamos utilizando inhibidores de moléculas pequeñas en modelos de ratón para dirigirnos a estas vÃas.
Tratamiento de células madre y progenitoras aberrantes en neoplasias mielodisplásicas:
Hemos demostrado que las células madre aberrantes persisten durante las remisiones morfológicas y están implicadas en las recaÃdas. Hemos identificado nuevos objetivos contra las células madre de SMD/LMA que se están probando en varios estudios. También estamos realizando análisis epigenéticos y de secuenciación de células madre humanas altamente purificadas y evaluando el papel de estas alteraciones en la transformación de SMD a LMA.
Análisis epigenómico de tumores:
Utilizamos ensayos de alto rendimiento para analizar la metilación del ADN en todo el genoma y hemos optimizado estos ensayos para que funcionen con pequeñas cantidades de ADN. Hemos utilizado con éxito estos ensayos para realizar un análisis integrador de alteraciones epigenéticas y genéticas en varios modelos tumorales, incluidos el sÃndrome mielodisplásico, el cáncer de esófago, el cáncer de páncreas y el cáncer de células renales.
