<p class="MsoNormal" style="text-align: justify;"><span style="font-size: 12.0pt; line-height: 107%; font-family: 'Times New Roman',serif;">Our lab studies biology and therapies for colorectal cancer. We use unique, patient-derived models of cancer in lab, and a combination of dry-lab and wet-lab approaches. Our techniques can be applied to mechanistic discovery as well as therapeutic validation of drugs. Ultimately, we hope to translate our findings into novel clinical trials.</span></p>
<p class="MsoNormal" style="text-align: justify;"><strong><span style="font-size: 12.0pt; line-height: 107%; font-family: 'Times New Roman',serif;">Inhibition of CDK9 for the treatment of CRC</span></strong><span style="font-size: 12.0pt; line-height: 107%; font-family: 'Times New Roman',serif;">: We have confirmed that novel small molecular inhibitors of Cyclin-dependent kinase 9 (CDK9) can potently inhibit CRC in our lab models. Our mechanistic work suggests that indirect suppression of EGFR/MAPK signaling is a major part of CDK9 inhibitor activity. Based on this finding, we hypothesized that inhibitors of the MAPK signaling pathway could be combined with CDK9 inhibitors to synergistically treat CRC. As proof-of-concept, we found that the BRAF inhibitor encorafenib indeed synergizes with CDK9 inhibitors to suppress CRC growth in vitro and in vivo. We are working to expand this project into other areas of MAPK targeting, as well as translate our findings clinically.</span></p>
<p class="MsoNormal" style="text-align: justify;"><strong><span style="font-size: 12.0pt; line-height: 107%; font-family: 'Times New Roman',serif;">Development of novel patient derived models for drug discovery</span></strong><span style="font-size: 12.0pt; line-height: 107%; font-family: 'Times New Roman',serif;">: Preclinical discovery and validation of cancer drugs is typically conducted using well-established cancer cell lines and xenograft models. These models are often monocultures devoid of clinically important features such as intra-tumoral heterogeneity and microenvironment. Further, the majority of established cancer models are derived from a homogeneous segment of the U.S. population. We are collecting cancer specimens from our diverse patients in the Montefiore Einstein Comprehensive Cancer Center to establish novel preclinical cancer models that will be used for cancer drug experiments. We will combine this biobank with rigorous molecular and clinical annotation to facilitate personalized drug discovery. To further extend our modeling capabilities, we are currently establishing more complex, co-culture models, in which non-carcinoma cells from the tumor microenvironment are cultured simultaneously with the carcinoma cells.</span></p>

<p>Chaoyuan Kuang, MD, PhD, is Assistant Professor, Oncology, Medicine and Molecular Pharmacology at Montefiore Einstein and Co-Leader, GI Clinical and Translational Research at Montefiore Einstein Cancer Center. His clinical focus is on the treatment of patients with colorectal and other gastrointestinal malignancies, as well as the enrollment of patients into clinical trials.</p><p>In 2007, Dr. Kuang graduated from the Massachusetts Institute of Technology, earning his Bachelor of Science in chemistry. He then attended the University of Michigan until 2015, earning his Doctor of Medicine as well as his Doctor of Philosophy in cell/molecular biology. For his postdoctoral training, Dr. Kuang went to the University of Pittsburgh Medical Center, completing an internship in medicine in 2016, a residency in medicine in 2017, a clinical fellowship in hematology/oncology in 2019 and a research fellowship in hematology/oncology in 2021.</p><p>Through preclinical research of colorectal cancer using tissue culture and mouse models of cancer, Dr. Kuang&rsquo;s research focus is on the discovery of novel therapies and biomarkers of cancer and scientific development of early phase trials for cancer. He has shared his research through peer-reviewed journals, book chapters, seminars, and oral and poster presentations.</p><p>Dr. Kuang is board certified in internal medicine. He is a member of several professional societies including the American Society for Clinical Oncology, the American Society of Hematology, the American Association for Cancer Research and the Society for Immunotherapy in Cancer.</p>

<p>Dennis L. Cooper, MD, is Chief, Bone Marrow Transplantation and Professor, Medicine at Montefiore Einstein. His clinical focus centers on treating patients with multiple myeloma, hodgkin's lymphoma, non-hodgkin's lymphoma including bone marrow transplantation and CAR T cell therapy.</p><p>After graduating from Loyola University of Chicago in 1975, Dr. Cooper completed his Doctor of Medicine at Rush Medical College in 1979. He then went to Yale-New Haven Hospital to complete his internship and residencies in internal medicine in 1982. Dr. Cooper became Chief Resident in internal medicine at the University of Pittsburgh in 1983, and then participated in a medical oncology fellowship at Yale-New Haven Hospital, completing it in 1985.</p><p>Dr. Cooper&rsquo;s research focuses on clinical trials for multiple myeloma and lymphoma and in the area of bone marrow transplant, he is interested in the use of alternative donor transplants (for patients who don&rsquo;t have a family match) as well as the impact of social determinants of health. Recently, he has published studies involving COVID-19 vaccine responses in patients with plasma cell dyscrasias after complete vaccination, persistent opioid use in patients with multiple myeloma post ASCT and posttransplant cyclophosphamide as part of standard immunoprophylaxis for all allogeneic transplants. His work has been published in many peer-reviewed journals and articles.</p><p>Dr. Cooper is a member of the American Society of Hematology and is board certified in Internal Medicine and Medical Oncology. He has won multiple awards, including the Yale Cancer Center Clinical Excellence award from Yale Cancer Center in 2010 and the Vida Award from the Leukemia and Lymphoma Society in 2018.</p>

<p><strong style="text-align: justify;">Targeting signal transduction in hematologic malignancies</strong><span style="text-align: justify;">:&nbsp;<br /></span><span style="text-align: justify;">Cytokines play important roles in the regulation of normal hematopoiesis and a balance between the actions of hematopoietic growth factors and myelosuppressive factors is required for optimal production of different hematopoietic cell lineages. The myelodysplastic syndromes (MDS) are collections of heterogeneous hematologic diseases characterized by refractory cytopenias due to ineffective hematopoiesis. These preleukemic disorders are common causes of anemia in the elderly and are rapidly increasing in incidence. We have also demonstrated that the TGF-beta superfamily/smad, IRAK and MAP kinase pathways is overactivated in MDS. Our work has led to clinical trials with Luspatercept, Galunisertib, CA4948, ARRY-614 and various other promising agents in MDS. We are trying to study the molecular mechanisms that lead to the activation of these pathways in MDS and are using small molecule inhibitors in mouse models to target these pathways.&nbsp;</span></p>
<p style="text-align: justify;"><strong>Targeting aberrant stem and progenitor cells in myelodysplastic neoplasms:&nbsp;<br /></strong>We have demonstrated that aberrant stem cells persist during morphological remissions and are involved in relapse. We have identified novel targets against MDS/AML stem cells that are being tested in various studies. We are also conducting sequencing/epigenetic analysis of highly purified human stem cells and evaluating the role of these alterations in transformation of MDS to AML.&nbsp;</p>
<p style="text-align: justify;"><strong>Epigenomic analysis of tumors</strong>: <br />We are using high throughput assays to analyze DNA methylation across the genome and have optimized these assays to work with low amounts of DNA. We have successfully used these assays in conducting an integrative analysis of epigenetic and genetic alterations in various tumor models, including MDS, esophageal cancer, pancreatic cancer and renal cell cancer.</p>
<p style="text-align: justify;"><strong>Clinical studies in Myelodysplastic syndromes</strong>: <br />We have a &ldquo;center of excellence&rdquo; clinic for patients with Myelodysplatic syndromes. We offer a variety of clinical trials for these patients.</p>

Dr. Verma conducts laboratory research aimed at discovering new treatments for myelodysplastic syndromes (MDS) and leukemias, and his research has shown that the TGF-beta superfamily/smad, IRAK and MAP kinase pathways are overactivated in MDS. Biography

<p>Amit K. Verma, MD, is Interim Chair, Department of Oncology, Associate Director of Translational Science, Montefiore Einstein Comprehensive Cancer Center, Professor, Medical Oncology and Professor, Developmental & Molecular Biology, Albert Einstein College of Medicine.</p><p>Dr. Verma earned his Doctor of Medicine at the All India Institute of Medical Sciences in 1995 and finished his training in internal medicine, hematology oncology and stem cell transplantation at the University of Illinois and Northwestern University in 2003.</p><p>Dr. Verma conducts clinical studies for patients with MDS and leukemia and offers a variety of clinical trials for these patients. He conducts laboratory research aimed at discovering new treatments for myelodysplastic syndromes (MDS) and leukemias, and his research has shown that the TGF-beta superfamily/smad, IRAK and MAP kinase pathways are overactivated in MDS. His work has led to clinical trials with Luspatercept, Galunisertib, CA4948, ARRY-614 and various other promising agents in MDS. DR. Verma&rsquo;s research has shown that MDS and leukemias are caused by aberrant leukemic stem cells and has identified novel targets against MDS/AML stem cells that are being tested in various studies.</p><p>Dr. Verma is a scholar of the Leukemia and Lymphoma society, elected member of American society of clinical investigation and his research is funded by the NIH, Department of Defense, American Cancer Society, Edward P. Evans Foundation and various other private foundations.</p>
