<p class="MsoNormal">My laboratory and clinical research are focusing on novel agents in acute myeloid leukemia. The main area of active investigations is attacking apoptotic machinery of leukemia cells by depriving them from key pro-survival inputs by targeting BCL-2, BCL-XL, Mcl-1 by small molecule inhibitors and degraders. We continue studies characterizing mechanisms of resistance to BCL-2 inhibition and identifying novel combinatorial strategies with immune therapies and signaling inhibitors. My lab is interested in studying the biology and therapeutic targeting of leukemia-initiating cells, with novel multi-targeted bispecific T- or NK-engagers, antibody-drug conjugates and CART cells. We further aim at identifying key targets in maintenance and eradication of minimal residual disease (MRD) in acute myeloid leukemia (AML). We have studied metabolic states in AML and T-ALL and utilized novel metabolic inhibitors to selectively deprive tumor metabolism, such as glutaminase, OxPhos and fatty acid metabolic inhibitors. Finally, we are interested in understanding the biology and targeting of an aggressive hematologic malignancy Blastic Plasmacytoid Dendritic ell neoplasm (BPDCN), through targeting BCL-2, CD123 and novel cell surface targets with small molecules, antibodies and CART cells. Most robust pre-clinical discoveries have translated into clinical trials, such as venetoclax, metabolic inhibitors and immune modulators.</p>

<p>Marina Konopleva, MD, Phd, is Professor, Oncology, Professor, Molecular Pharmacology, Director, Leukemia Program and Co-Director, Blood Cancer Institute, at Montefiore Einstein. Her clinic focus centers on treating patients with hematologic malignancies, with an emphasis on acute myeloid and acute lymphoblastic leukemia. She has additional expertise in treating patients with high-risk myelodysplastic syndrome and chronic myeloid leukemia.</p><p>After obtaining her Doctor of Medicine in 1990 from First Pavlov Medical Institute in St. Petersburg, Russia, Dr. Konopleva completed her postgraduate training in internal medicine at I.P. Pavlov State Medical Institute in 1993. In 1998 she obtained her PhD in experimental hematology from the Federal Institute of Hematology and Blood Transfusion as well as her postdoctoral fellowship in Molecular Hematology with The University of Texas MD Anderson Cancer Center.</p><p>Dr. Konopleva conducts laboratory research focusing on approaches activating cell death, metabolic inhibitors and immune-oncology in AML. Her key contributions include the identification of dependency of AML cells on the anti-apoptotic protein BCL-2 and the FDA approval of the BCL-2 inhibitor venetoclax in combination with low-intensity chemotherapy in elderly patients unfit for standard chemotherapy. This therapy provides both low toxicity for elderly patients with AML and documented survival benefit. Dr. Konopleva&rsquo;s ground-breaking research greatly benefited leukemia research, clinical research and the lives of patients. She is a scholar of the Leukemia and Lymphoma Society and her research is funded by the NIH, Department of Defense and various private foundations. Her work has been published in numerous peer-reviewed journals and she has presented both nationally and internationally.</p><p>Dr. Konopleva is board certified by the Russian Board of Hematology, the American Board of Internal Medicine and the American Board of Internal Medicine - Hematology. She is a member of numerous professional societies including the American Society of Hematology, the American Association of Cancer Research, the American Society of Clinical Oncology, the American Society for Clinical Investigation and the Society of Hematologic Oncology.</p><p>Dr. Konopleva has won many awards for her work, such as the R. Lee Clark Prize - Basic/Translational Science and the Students' Faculty Choice Award from MD Anderson Cancer Center in 2019, the Otis W. and Pearl L. <p>Carolina Bernabe, MD, is a Medical Gastrointestinal Oncologist and Assistant Professor at Montefiore Einstein. Her clinical focus is in the treatment of colorectal cancer and gastrointestinal malignancies.</p><p>After earning her Doctor of Medicine at Universidad Cat&oacute;lica Santiago de Guayaquil in Guayaquil, Ecuador in 2010, Dr. Bernabe completed her internal medicine residency at Mount Sinai Beth Israel in 2016. <p>Adilia Hormigo, MD, PhD, is Leader, Neuro-Oncology Program, Co-Leader, Neuro-Oncology Multidisciplinary Disease Team and Professor, Hematology-Oncology, Neurosurgery, and Microbiology &amp; Immunology at Montefiore Einstein. She is also a member of the Cancer Therapeutics Program. Her clinical expertise is the treatment of malignancies of the central nervous system and neurologic complications of cancer.</p><p>Dr. Hormigo earned her Doctor of Medicine with honors from the University of Lisbon and her Doctor of Philosophy from New York University. She pursued a residency in neurology at New York Presbyterian-Weill Cornell University/Memorial Sloan-Kettering Cancer Center. She also completed research and clinical neuro-oncology fellowships at Memorial Sloan-Kettering Cancer Center. <p>Dennis L. Cooper, MD, is Chief, Bone Marrow Transplantation and Professor, Medicine at Montefiore Einstein. His clinical focus centers on treating patients with multiple myeloma, hodgkin's lymphoma, non-hodgkin's lymphoma including bone marrow transplantation and CAR T cell therapy.</p><p>After graduating from Loyola University of Chicago in 1975, Dr. Cooper completed his Doctor of Medicine at Rush Medical College in 1979. He then went to Yale-New Haven Hospital to complete his internship and residencies in internal medicine in 1982. Dr. Cooper became Chief Resident in internal medicine at the University of Pittsburgh in 1983, and then participated in a medical oncology fellowship at Yale-New Haven Hospital, completing it in 1985.</p><p>Dr. Cooper&rsquo;s research focuses on clinical trials for multiple myeloma and lymphoma and in the area of bone marrow transplant, he is interested in the use of alternative donor transplants (for patients who don&rsquo;t have a family match) as well as the impact of social determinants of health. Recently, he has published studies involving COVID-19 vaccine responses in patients with plasma cell dyscrasias after complete vaccination, persistent opioid use in patients with multiple myeloma post ASCT and posttransplant cyclophosphamide as part of standard immunoprophylaxis for all allogeneic transplants. His work has been published in many peer-reviewed journals and articles.</p><p>Dr. Cooper is a member of the American Society of Hematology and is board certified in Internal Medicine and Medical Oncology. He has won multiple awards, including the Yale Cancer Center Clinical Excellence award from Yale Cancer Center in 2010 and the Vida Award from the Leukemia and Lymphoma Society in 2018.</p>

<p><span>Dr. Allan Wolkoff's interest in research was sparked when he worked in a biochemistry lab to help pay college bills. He began his medical studies at Dartmouth Medical College and transferred to the Albert Einstein College of Medicine where he obtained his M.D. degree. <p><strong style="text-align: justify;">Targeting signal transduction in hematologic malignancies</strong><span style="text-align: justify;">:&nbsp;<br /></span><span style="text-align: justify;">Cytokines play important roles in the regulation of normal hematopoiesis and a balance between the actions of hematopoietic growth factors and myelosuppressive factors is required for optimal production of different hematopoietic cell lineages. The myelodysplastic syndromes (MDS) are collections of heterogeneous hematologic diseases characterized by refractory cytopenias due to ineffective hematopoiesis. These preleukemic disorders are common causes of anemia in the elderly and are rapidly increasing in incidence. We have also demonstrated that the TGF-beta superfamily/smad, IRAK and MAP kinase pathways is overactivated in MDS. Our work has led to clinical trials with Luspatercept, Galunisertib, CA4948, ARRY-614 and various other promising agents in MDS. We are trying to study the molecular mechanisms that lead to the activation of these pathways in MDS and are using small molecule inhibitors in mouse models to target these pathways.&nbsp;</span></p>
<p style="text-align: justify;"><strong>Targeting aberrant stem and progenitor cells in myelodysplastic neoplasms:&nbsp;<br /></strong>We have demonstrated that aberrant stem cells persist during morphological remissions and are involved in relapse. We have identified novel targets against MDS/AML stem cells that are being tested in various studies. We are also conducting sequencing/epigenetic analysis of highly purified human stem cells and evaluating the role of these alterations in transformation of MDS to AML.&nbsp;</p>
<p style="text-align: justify;"><strong>Epigenomic analysis of tumors</strong>: <br />We are using high throughput assays to analyze DNA methylation across the genome and have optimized these assays to work with low amounts of DNA. We have successfully used these assays in conducting an integrative analysis of epigenetic and genetic alterations in various tumor models, including MDS, esophageal cancer, pancreatic cancer and renal cell cancer.</p>
<p style="text-align: justify;"><strong>Clinical studies in Myelodysplastic syndromes</strong>: <br />We have a &ldquo;center of excellence&rdquo; clinic for patients with Myelodysplatic syndromes. We offer a variety of clinical trials for these patients.</p>

Myelodysplastic syndrome (MDS) are hematologic malignancies characterized by ineffective hematopoiesis. We study the molecular pathogenesis of MDS to identify targetable pathways for treatment of this disease.

Research Focus

Dr. Verma conducts laboratory research aimed at discovering new treatments for myelodysplastic syndromes (MDS) and leukemias, and his research has shown that the TGF-beta superfamily/smad, IRAK and MAP kinase pathways are overactivated in MDS. His work has led to clinical trials with Luspatercept, Galunisertib, CA4948, ARRY-614 and various other promising agents in MDS. DR. Verma’s research has shown that MDS and leukemias are caused by aberrant leukemic stem cells and has identified novel targets against MDS/AML stem cells that are being tested in various studies.

<p>Amit K. Verma, MD, is Interim Chair, Department of Oncology, Associate Director of Translational Science, Montefiore Einstein Comprehensive Cancer Center, Professor, Medical Oncology and Professor, Developmental & Molecular Biology, Albert Einstein College of Medicine.</p><p>Dr. Verma earned his Doctor of Medicine at the All India Institute of Medical Sciences in 1995 and finished his training in internal medicine, hematology oncology and stem cell transplantation at the University of Illinois and Northwestern University in 2003.</p><p>Dr. Verma conducts clinical studies for patients with MDS and leukemia and offers a variety of clinical trials for these patients. He conducts laboratory research aimed at discovering new treatments for myelodysplastic syndromes (MDS) and leukemias, and his research has shown that the TGF-beta superfamily/smad, IRAK and MAP kinase pathways are overactivated in MDS. His work has led to clinical trials with Luspatercept, Galunisertib, CA4948, ARRY-614 and various other promising agents in MDS. DR. Verma&rsquo;s research has shown that MDS and leukemias are caused by aberrant leukemic stem cells and has identified novel targets against MDS/AML stem cells that are being tested in various studies.</p><p>Dr. Verma is a scholar of the Leukemia and Lymphoma society, elected member of American society of clinical investigation and his research is funded by the NIH, Department of Defense, American Cancer Society, Edward P. Evans Foundation and various other private foundations.</p>

<p>Roberto Alejandro Sica, MD, is an Attending Physician at Montefiore and an Assistant Professor of Medicine at our Albert Einstein College of Medicine. Since joining the Montefiore team, Dr. Sica&rsquo;s clinical focus has been on chimeric antigen receptor (CAR) T-cell therapies, immunotherapies and allogeneic stem cell transplantation, central nervous system (CNS) lymphomas and HIV, ATLL and gene therapy for malignant and nonmalignant hematologic disorders. Dr. Sica is a member of the Bone Marrow Transplant team at Montefiore.</p><p>Dr. Sica attended the University of Buenos Aires School of Medicine, where he received his Medical Degree. During his time there, he also served as an instructor of immunology, virology, pharmacology, and physical diagnosis. Dr. Sica began his postgraduate training with a residency in Internal Medicine at the Carney Hospital/Tufts University School of Medicine in Boston. Following this, he completed a fellowship in Hematology/Oncology at the University of Illinois at Chicago, where he received the E. Donnall Thomas Achievement in Medicine Award for his academic performance and the Edward Jenner Innovation in Medicine Award in recognition of his innovative contributions to the design of clinical protocols and trials. Later, he completed a fellowship in Blood and Marrow Transplant and Cellular Therapies at Stanford University. At that institution, he performed research with Dr. Sally Arai on Hodgkin's lymphoma using the Stanford-pioneered GN-BVC regimen (Gemcitabine, Vinorelbine-BCNU. Etoposide and Cyclophosphamide) which resulted in decreased toxicity and improved overall survival in patients. His work has been published in various review journals and abstracts.</p><p>Dr. Sica&rsquo;s research focuses on novel cellular therapies, allogeneic bone marrow transplantation for malignant and nonmalignant hematologic disorders, and combination immunotherapy and cancer vaccine.</p><p>Dr. Sica is board certified in Internal Medicine, Hematology and Medical Oncology by the American Board of Internal Medicine.</p>

Biography

<p>Nishi Shah, MD, MPH, is an attending physician, Oncology, and Assistant Professor at Montefiore-Einstein. Her clinical focus is on the treatment of patients with hematologic malignancies, with particular focus in plasma cell disorders and B-cell malignancies. She utilizes novel targeted agents, stem cell transplant and cell therapies in the treatment of patients.
</p><p>After earning her medical degree at Grand Medical College in Mumbai, India in 2011, Dr. Shah attended the Rollins School of Public Health at Emory University, earning her Master of Public Health in 2014. She then pursued postgraduate training at University of Arkansas for Medical Sciences, completing her internal medicine internship in 2015 and her internal medicine residency in 2017. She then came to Montefiore to pursue a hematology-oncology fellowship, completing it in 2020.
</p><p>Dr. Shah&rsquo;s research focuses on improving access to care and addressing gaps in health equity for patients with hematologic malignancies as well as exploring methods to improve patient access to clinical trials. She has shared her work through peer-reviewed journals, abstracts, and poster presentations. Dr. Shah is also an ad hoc reviewer for several journals including Acta Hematologica, Leukemia and Lymphoma, and Clinical Lymphoma, Myeloma and Leukemia.
