Cortex-basal ganglia interactions in drug addiction and motivated behavior

Drug addiction and other neuropsychiatric conditions involve complex interactions between brain areas including the nucleus accumbens, hippocampus, and prefrontal cortex. Our <a href="http://sjulsonlab.org/wp-content/uploads/2018/06/1-s2.0-S08966273183032…; target="_blank" rel="noopener noreferrer">previous work has characterized the role of interactions between hippocampus and nucleus accumbens in cocaine conditioned place preference, finding evidence that selective potentiation of specific hippocampal inputs to accumbens stores information linking certain spatial contexts to drug use. This provides a possible mechanistic substrate for the well-known phenomenon by which relapse is triggered by exposure to “people, places, and things” previously associated with drug use.

<a href="http://sjulsonlab.org/research/" target="_blank" rel="noopener noreferrer">Our current work studies how interactions between hippocampus and accumbens influence prefrontal cortex as a basis for both physiological reward valuation and the pathological overvaluation of drugs of abuse. To this end, we combine carefully-designed mouse behavioral paradigms with a broad range of experimental techniques including multisite Neuropixels probe recordings, optotagging and closed-loop optogenetic manipulations, in vivo two photon and light field imaging, and in situ RNA sequencing. A major theme of this work is linking transcriptomically-defined cell classes to their functional roles during behavior. We also rely heavily on dimensionality reduction approaches, machine learning, and computational modeling to characterize and understand the population… coordination of neuronal activity between brain regions during decision making and drug self-administration.

Developing novel translational strategies for treating drug addiction

Drug addiction represents a large public health burden for which few effective biologically-based treatments exist. We aim to develop novel gene-based tools and validate anatomical targets for future translational studies in human subjects. Our <a href="http://sjulsonlab.org/wp-content/uploads/2018/02/Cassataro2014.pdf" target="_blank" rel="noopener noreferrer">previous work in this area has focused on using <a href="https://en.wikipedia.org/wiki/Receptor_activated_solely_by_a_synthetic_…; target="_blank" rel="noopener noreferrer">DREADDs to modulate nucleus accumbens and suppress alcohol consumption in a model of binge drinking. We are currently expanding this work by exploring different brain targets and developing novel DREADD-like receptors with properties suitable for use in human subjects. We have also developed several novel molecular tools enabling manipulation of synaptic transmission and plasticity with greater precision than previously possible.

Highlighted awards

Dr. Sjulson was the recipient of a five-year, $2.5 million National Institutes of Health Director's Pioneer Award from the National Institute on Drug Abuse (NIDA).

Dr. Shulman serves as the director of Infant Toddler Services at Montefiore’s Children’s Evaluation and Rehabilitation Center (CERC), which provides family-based diagnostic assessment and treatment services for young children who have or are suspected of having developmental delays. She also directs CERC’s RELATE program, which offers evaluation and treatment for young children with autism and ASD.

Her research interests include early identification of autism, healthcare disparity in autism diagnosis and management, and the use of complementary and alternative medicine in treating autism.

Dr. Shulman recently led a study that characterized the residual learning and cognitive needs of school-aged children whose early ASD diagnosis resolved. The study showed that most children who outgrow their autism diagnosis still struggle with emotional, behavioral or leaning difficulties later in life.

Biography

Lisa Heidi Shulman, MD, is Professor of Pediatrics at Albert Einstein College of Medicine. Since joining the Montefiore team, her clinical focus has been on the early identification and long-term management of children with autism spectrum disorder (ASD), as well as improving access to care for our community, identification and management of early motor delays and toe walking gait.

In 1983, Dr. Shulman received a Bachelor of Science in Applied Mathematics/Psychology at Brown University. She then attended the University of Pennsylvania School of Medicine, where she received her Doctor of Medicine in 1987. Dr. Shulman began her postgraduate training in 1987 with a 3-year internship and residency in Pediatrics at Mount Sinai Medical Center. In 1992, she completed a clinical fellowship in Pediatric Neurodevelopment at the Rose F. Kennedy Center, Children’s Evaluation and Rehabilitation Center at Albert Einstein College of Medicine.

Aligning with her clinical interests, her research also focuses on other aspects of ASD, including early diagnosis, developmental outcomes in children with an early diagnosis, comorbid medical and developmental problems, parental stress and complementary and alternative medicine usage in ASD. Dr. Shulman is involved in numerous projects involving ASD, including one that began in 2019 that aims to identify the causes of ASD by carrying out genetic testing, phenotyping, and demographic profiles on the parents of children with ASD. Her work has been shared nationally at presentations and lectures and has been published in a number of peer-reviewed journals, books, review articles and abstracts. She is a featured contributor for Einstein’s blog, the Doctor’s Tablet (https://blogs.einsteinmed.edu/</a>) and has produced popular videos demonstrating baby milestones (https://www.youtube.com/wa… 2008 to 2019, Dr. Shulman has been named one of “New York’s Super Doctors: Best Physicians as Chosen by Their Peers.” She served as the Centers for Disease Control Act Early Ambassador to New York State from 2016-2019.
